It used to be a bit more difficult to get to the slot canyon, but as of Winter 2013, the dirt road has been re-graveled, and a dip just before the wash (which wasn't passable before) can now be driven down.
It's very possible to drive all the way up to the canyon itself, but I highly suggest parking at the bottom of the wash and walking the short 15 minute hike. The spot is quite popular for hikers, and having cars parked along the traditional hiking route can spoil the feel of a hike. Be courteous, the hike's not hard.
DIRECTIONS
Leave Las Vegas and head towards Lake Mead. Once in the park, take the first left along Lakeshore Rd. If you entered on Lake Mead Parkway in Henderson, you'll be driving 15.8 miles to Callville Wash Rd. If you enter the park from Lake Mead Boulevard in North Las Vegas, it's 12.6 miles.
Once you've taken the small turnoff to the left, the dirt road portion begins. It's not a bad road, and I've driven it in a 2004 Chevy Impala and a 2012 Chevy Cruze without ANY issues, it's a decent, well traveled dirt road. There's a left turn about 100 yards in (and the obvious route, it's much more developed.
There's another fork about 1.3 miles down and you'll follow that to the left as well. Keep straight as it turns right after another half mile, and .25 miles later you're in the wash. I recommend parking here, and walking the rest of the way, just follow the wash North, and you'll be at the canyon opening in 15 minutes or so (it is .75 miles, and it's all pretty much flat.)
Here's the route of the dirt road / walking portion. Essentially, stay to the left along the dirt road until you get into the wash, and then walk north. Click for a larger version. The white dot with "Narrows" next to it is where you should park.
Something awesome, that makes it super easy --- if you've got Google Maps Navigator on your phone, you can literally just say "Anniversary Mine" and it will give you step by step directions all the way to the mouth of the narrows, it's pretty brilliant.
